Sterility Test of Clindamycin Hydrochloride for Injection

Objective To improve the sterility test of clindamycin hydrochloride for injection. Methods By determining the amount of dilution and flushing volume,the positive control was selected. The clindamycin hydrochloride for injection (0.6 g) was used as an example to show the necessity of sterility test based on a basis of verification test. Results The verification test of clindamycin hydrochloride for injection could ensure the scientific character of sterility test and the accuracy test results. Conclusion The result can further improve the sterile test methods in Chinese Pharmacopoeia (Edition 2005,Vol II).

Effects of clindamycin hydrochloride on secretion of TNF-α in LPS-induced human periodontal ligament cells

Objective To explore the effect of clindamycin hydrochloride(CA)on lipopolysaccharide(LPS)-induced human periodontal ligament cells(HPDLCs)secreting tumor necrosis factor-α(TNF-α). Methods Primary cultured HPDLCs were randomized into 6 groups:blank group,LPS group and CA groups.HPDLCs were added 100 μg/ml LPS in LPS group,while LPS(100 μg/ml)and diffe-rent concentrations of CA(2.5,10,40,160 μg/ml)were added in CA groups,respectively.Enzyme-linked immunosorbent assay method(ELISA)was used to detect the level of TNF-α. Results The level of TNF-α in LPS group was significantly higher than in blank group.TNF-α levels were lower in CA groups than in LPS group,especially in 10,40,160 μg/ml CA groups(P0.05),but there was no significant difference between CA groups(P0.05). Conclusion LPS can promote the secretion of TNF-α.CA in concentration of 10,40,60 μg/ml can inhibit secretion of TNF-α in LPS-induced human periodontal ligament cells,which may be one of anti-inflammatory mechanisms.
